Intensified biomass harvesting could prove to be negative for forest ecological health through the impacts this type of forest management could exert on microbial community structure and function in forest soils and in CWD pools. Microbial community functional characteristics as well as community structure (through T-RFLP and pyrotag sequencing of ssu rRNA) were assayed soils in a boreal jack pine forest exposed to a clearcut intensified harvesting gradient. Microbial communities within CWD of various decay stages were also assessed in order to determine habitat specificity of the decomposer communities within them. Soil microbial communities were altered by harvesting, but intensification did not cause further disturbance. Soils in harvested sites were different from fire sites also assayed, meaning that these disturbance types may have different impacts on microbial community structure and functioning. CWD communities within logs had different characteristics in different sites. Intensification could reduce site specific organisms important in decay initiation.

Research on decomposer communities of terrestrial ecosystems for a long time has focussed on microbial biomass and gross turnover parameters. Recently, more and more attempts are made to look beyond the biomass, and more specifically determine functions and populations on a smaller scale-in time and space. A multitude of techniques is being improved and developed. Garland and Mills (1991) triggered a series of publications on substrate utilization tests in the field of microbial ecology. Despite several promising results for different applications in different laboratories, many problems concerning the assay and the interpretation of results became evident. After individual discussions on the approach with colleagues from various laboratories we started to plan a workshop on the matter. The response on our first circular was extraordinary, and instead of a small workshop it became a meeting with almost 150 participants. The meeting was named 'Substrate use for characterization of microbial communities in terrestrial ecosystems' (SUBMECO) and was held in Innsbruck, Austria, from Oct. 16-18, 1996. The very focussed scope attracted enthusiastic advocates of the approach, and also serious critics. Some of the topics concerned improvements of current inoculation and incubation techniques, ranging from sample pre-treatment, inoculum density and incubation temperature to statistical data handling. New methods for calculating microbial diversity were proposed, as well as bootstrap methods that allow statistics with many variables on a relatively low number of replicates.

A more complete understanding of the effects of intensive forest biomass harvesting on the soil microbial community, soil nutrient bioavailability, and soil organic carbon (SOC) dynamics are essential to advance knowledge of maintaining forest productivity under sustainable biomass harvesting. The objectives of this research are: (1) to quantify the effects of different harvesting practices on shifts in the soil microbial community (2) to address the long-term effects of soil compaction on the soil microbial community and soil nutrients in surface (0-10 cm) and subsurface (20-30 cm) mineral soil two decades after harvest; (3) to determine the effects of organic matter and woody biomass removal combined with soil compaction on SOC pools and mean residence time (MRT) in surface and subsurface mineral soil two decades after harvest, and relate these effects to soil biotic and abiotic C stabilization mechanisms. Four established field experiments were used to assess the effects of intensive biomass harvests on ecosystem properties. The research sites are located throughout the central lake states (MI, WI, MN), in aspen and northern hardwood forests. Soil microbial community composition and biomass were quantified using phospholipid fatty acid (PLFA) analysis. Soil nutrient bioavailability was assessed using PRS nutrient probes. SOC dynamics were assessed using a density fractionation procedure followed by carbon and nitrogen determination. Information from this research will help us to address the goal of balancing our global need for energy with sustainable forest use.
