Redox-Active Molecules as Therapeutic Agents available in PDF, EPUB and Kindle. Oxidative stress and altered redox signaling have been described in a plethora of pathological conditions. Redox-active molecules can thus potentially be used to modulate the etiology/progression of such diseases. Recent advances in molecular biology and pharmacology have strengthened this area of research by providing novel mechanistic insights. This book compiles a collection of 13 articles, covering a range of topics from in vitro studies to clinical research, focused on the potential therapeutic effects of either natural or synthetic compounds, applicable to different redox-related diseases.

This essential volume comprehensively discusses redox-active therapeutics, focusing particularly on their molecular design, mechanistic, pharmacological and medicinal aspects. The first section of the book describes the basic aspects of the chemistry and biology of redox-active drugs and includes a brief overview of the redox-based pathways involved in cancer and the medical aspects of redox-active drugs, assuming little in the way of prior knowledge. Subsequent sections and chapters describe more specialized aspects of central nervous system injuries, neurodegenerative diseases, pain, radiation injury and radioprotection (such as of brain, lungs, head and neck and erectile function) and neglected diseases (e.g., leishmaniasis). It encompasses several major classes of redox-active experimental therapeutics, which include porphyrins, salens, nitrones, and most notably metal-containing (e.g., Mn, Fe, Cu, Zn, Sb) drugs as either single compounds or formulations with nanomaterials and quantum dots. Numerous illustrations, tables and figures enhance and complement the text; extensive references to relevant literature are also included. Redox-Active Therapeutics is an invaluable addition to Springer’s Oxidative Stress in Applied Basic Research and Clinical Practice series. It is essential reading for researchers, clinicians and graduate students interested in understanding and exploring the Redoxome—the organism redox network—as an emerging frontier in drug design, redox biology and medicine.

Synthesis of Redox Active Neuroprotective Therapeutic Agents available in PDF, EPUB and Kindle. Mitochondria are energy-producing organelles present in eukaryotic cells. Energy as adenosine triphosphate (ATP) is produced at the end of a series of electron transfers called the electron transport chain (ETC). Such a highly coordinated and regulated series of electron transfer reactions give rise to a small percentage of electron leakage which, by the subsequent reduction of molecular oxygen, produce superoxide anions (O2.-). These anions initiate the production of additional highly reactive oxygen-containing radicals commonly known as reactive oxygen species (ROS). Although cells are equipped with endogenous antioxidant systems to minimize ROS accumulation, these endogenous defense systems become inadequate when ROS generation is increased. When ROS production occurs in excess, the cell is said to be under oxidative stress. Unchecked ROS production causes damage to cellular macromolecules, which in turn leads to cell death. Dysfunctional mitochondria and subsequent cell degeneration are a common cause of neurodegenerative diseases such as Friedreich’s ataxia (FRDA) and Alzheimer’s disease (AD). Therefore, targeting the mitochondria by neuroprotective drugs is imperative for the treatment of such diseases. In Chapter 1, the functioning of the ETC is described. Moreover, excessive ROS production and its consequences are also described. FRDA is a progressive neurodegenerative disease caused by insufficient expression of frataxin (FXN). FXN is instrumental in the assembly of iron-sulfur clusters, which in turn are critical for the functioning of the ETC enzyme complexes. Therapeutic agents which, in addition to being antioxidants also increase FXN, can be good drugs to counter FRDA. In Chapter 2, the synthesis of phenothiazine analogues are described. Moreover, their efficacy as antioxidants and their ability to increase FXN are described. Finally, the synthesis of a reduced salt form of one analogue and its ability to cross the blood brain barrier (BBB) in mouse models of the disease is also described. In Chapter 3, to discover potent neuroprotective drugs, a pair of regioisomeric benzoquinone analogues has been synthesized. The compounds were tested for their efficacy as antioxidants. Additionally, two pyrimidinol based redox cores were analyzed electrochemically to enable a better understanding of the mechanism of action of the multifunctional radical quencher (MRQ) class of antioxidants.

Recent Advances in Redox Active Plant and Microbial Products available in PDF, EPUB and Kindle. Nature endows us with a treasure chest of Green Gold full of amazing ‘redox-active’ substances which interfere with numerous biological processes in our own body, in animals, bacteria, fungi and plants. Whilst such natural products are all around and also in us, we still do not fully understand how these compounds actually work. This book attempts to resolve some of the mysteries and riddles associated with such products. Written by more than thirty international experts from academia and industry, it places a focus on modern developments in this field and considers such natural products from various angles, from their isolation and characterization all along to product development and commercialization. Throughout, the reader will be confronted with modern approaches which enable the efficient identification and isolation of new natural products, help to elucidate their mode(s) of action and permit practical uses in Medicine, Cosmetics, Agriculture, Industry and as functional foods.
